Post DOE Robust Simulation. Topics, Techniques and Tools

A post-DoE technique, which would give further added value to DoE, is the Monte Carlo simulation that uses the obtained models, with the target to better manage the solutions in a long-term industrial process.

Paul Nelson

PhD, Technical Director, Prism Training & Consulting, Ltd.

Practical (Real-life) Implementation of Definitive Screening Designs

Custom Designs in Design-Expert® implement the idea of model-oriented design. That is, they maximise the information about a specified model. This suggests the analysis of designed experiments profits by taking advantage of what is already known about their structure: design-oriented modelling.

Philippe Pages

Independent Consultant & Trainer, PhP Stats

How to Evaluate the Quality of an Evolving Design?

Several saturated designs in 22 runs do exist: Min-Run Characterize (Design Expert), Rechtschaffner matrix (literature)… 22 being not a power of two they cannot be perfectly orthogonal. Which is the best? In addition, initially planned factor levels cannot always be respected. What is the impact of those variations on the design quality?

Dimitris Alexandrakis

Business Development Director CAMO Software AS

Design of Experiments and Multivariate Analysis - a symbiotic partnership

The presentation will show various approaches to analyse data and visualise results from experimental designs in the case of multiple responses. An example with observed variables as covariates for a multivariate alternative MANCOVA will also be discussed.

B. Thomas Carr

Principal, Carr Consulting

Modeling Sweet-Taste Synergy in Blends of High Potency Sweeteners Using Mixture Design

Synergy has substantial economic impact for manufacturers who use high potency sweeteners in their products. Identifying the ratio of sweeteners that maximize sweet-taste synergy offers substantial cost savings opportunities. A three-component mixture experiment was developed using "Design-Expert" to determine the proportions of Aspartame, Acesulfame-K, and Neotame that maximize sweet-taste synergy.

David Brian Nicolaides

Solutions Consultant, Dassualt Systemes BIOVIA

Better Management of Computer Experiments Using Design-Expert

We discuss a particular application in the domain of chemical reactions, where we want to minimize the reaction byproducts by understanding and optimizing the reaction kinetics (which are notoriously difficult to measure). Design Expert together with Modelica, an open-source dynamic simulation toolkit, has been used to find the optimum reaction parameters, with quantifiable benefits for the reaction yield.

Francesco Tinazzi

Senior Scientist, Aptuit Verona

Use of DOE to Increase Process Understanding of a De-Bromination Reaction

The presentation describes the use of DoE to optimize and increase the process understanding of a de-bromination reaction via catalytic transfer hydrogenation (CTH), a key step in the synthesis of a potential drug, using formic acid as hydrogen donor and 5% palladium on charcoal as catalyst.

Daniel Tray

Scientific Investigator, API Chemistry, GSK

The Role of Fractional and D-Optimal Designs in the Development of a QbD Pharmaceutical Production Process: A Process Chemistry Case Study

This talk will exemplify how DoEs created by "Design-Expert" software have been used to explore multiple unit operations of a complex chemical process and deliver the understanding necessary to comply with the requirements of a Quality-by-Design environment. These DoEs encompassed both traditional fractional factorial designs as well as resource efficient non-orthogonal D-Optimal designs. The challenges associated with analysing a sparse data set will be discussed and model selection strategies will be presented.

Christos Georgakis

Professor, Tufts University & Process Cybernetics LLC

DoDE & DRSM: Two Generalizations of the DoE and RSM Methodologies

This talk presents two generalizations Design of Dynamic Experiments (DoDE) and Dynamic Response Surface Methodology (DRSM). The first, DoDE, enables the design of experiments with time-varying inputs something that can be easily achieved with Design Expert. The Second one (DRSM) enables the development of RSM-type models when process measurements are available at several time instants during the experiment

Transferring a Drug Product Formulation Design Space from Development to Production—Some Hurdles with DOE Application

Extended release (ER) formulations, and particularly granules, are commonly used as drug products. They have the advantage of releasing the Active Pharmaceutical Ingredient (API) over a long period providing the patient with a long-lasting therapeutic effect.

Johannes Felix Buyel

Design of Experiments Facilitate Optimization in Biopharmaceutical Protein Screening—a Case Sudy

In order to identify highly potent, safe and affordable biopharmaceuticals, an immense screening effort is required. We have therefore used a design of experiments (DoE) approach to identify the most relevant factors and to select conditions facilitating reproducible outcomes which is a prerequisite to use the system for subsequent high-throughput screening applications.

  Tim Revett

Science lead in Cell Culture Development at  Lonza Biologics

Design of experiment in Cell Culture Development and Process Development Sciences

A custom I-optimal response surface model was used to evaluate the effects of six factors in the process, with the aim to reduce the concentration of the increased metabolite,  whilst maintaining a high culture viability and productivity.  A high through-put, miniature bioreactor system was used to evaluate the model and generated a set of optimised conditions that met these requirements.

Markus Höfer

Development and Optimization of highly transparent SiO2 films

The challenge was to find process parameters offering the opportunity to apply sufficiently high
amounts of oxygen, in order to make the SiO2 films completely stoichiometric, without
significant evaporation of tungsten oxides from the wires. By using DOE we succeeded, for the
first time ever, to develop a stable HWCVD process for preparing highly transparent SiO2 films
with almost no tungsten contaminants.

New Peel Seal System for Flexible Medical Device Packaging Applications

A newly developed uncoated nylon reinforced paper provides an alternative to materials traditionally used for lidding. This paper serves to demonstrate the successful use of a Mixture DOE in the development of a new peel/seal system.

Ashraf Zin

Senior scientist with PETRONAS

A statistical based study employing Response Surface Method (RSM) was conducted to ascertain the optimum lignocellulosic pretreatment conditions i.e. at different temperature, time and base concentration with Malaysian oil palm empty fruit bunches (EFB) was used as a model substrate.

Practical DOE “Tricks of the Trade” by Pat Whitcomb and Mark Anderson
